چکیده

The genus Streptococcus is one of the most genomically diverse and important human and agricultural pathogens. The acquisition of genomic islands (GIs) plays a central role in adaptation to new hosts in the genus pathogens. The research presented here employs a comparative genomics approach to define a novel family of GIs in the genus Streptococcus which also appears across strains of the same species. Specifically, we identified 9 Streptococcus genomes out of 67 sequenced genomes analyzed, and we termed these as 15bp Streptococcus genomic islands, or 15SGIs, including i) insertion adjacent to the 3’ end of ribosome l7/l12 gene, ii) large inserts of horizontally acquired DNA, and iii) the presence of mobility genes (integrase) and replication initiators. We have identified a novel family of 15SGIs and seems to be important in species differentiation and adaptation to new hosts. It plays an important role during strain evolution in the genus Streptococcus. peer-reviewed) is the author/funder. All rights reserved. No reuse allowed without permission. Background: Staphylococcus aureus and Staphylococcus epidermidis are major human pathogens of increasing importance due to the spread of antibiotic resistance. Novel potential targets for therapeutic antibodies are products of staphylococcal genes expressed during human infection. Previously, the secreted and surface-exposed proteins among seroreactive antigens have been discovered. Furthermore...

New insights in the molecular biology and physiology of Streptococcus thermophilus revealed by comparative genomics.

Streptococcus thermophilus is a major dairy starter used for the manufacture of yoghurt and cheese. The access to three genome sequences, comparative genomics and multilocus sequencing analyses suggests that this species recently emerged and is still undergoing a process of regressive evolution towards a specialised bacterium for growth in milk. Notably, S. thermophilus has maintained a well-de...

Biochemical characterization of PE_PGRS61 family protein of Mycobacterium tuberculosis H37Rv reveals the binding ability to fibronectin

Objective(s): The periodic binding of protein expressed by Mycobacterium tuberculosis H37Rv with the host cell receptor molecules i.e. fibronectin (Fn) is gaining significance because of its adhesive properties.  The genome sequencing of M. tuberculosis H37Rv revealed that the proline-glutamic (PE) proteins contain polymorphic GC-rich repetitive sequences (PGRS) which have clinical importance i...

Phylogeny of gazelles in some islands of Iran based on mtDNA sequences: Species identification and implications for conservation

Different species of gazelles are among the most endangered mammals on the Asian steppes and occur in the central, southern and northwestern regions of Iran. The previous conservation efforts in this region have been incomplete due to confusion about the phylogenetic relationship among various populations. So that, different conservation programs such as ex-situ breeding and transfer of captive...

Chromosomal islands of Streptococcus pyogenes and related streptococci: molecular switches for survival and virulence

Streptococcus pyogenes is a significant pathogen of humans, annually causing over 700,000,000 infections and 500,000 deaths. Virulence in S. pyogenes is closely linked to mobile genetic elements like phages and chromosomal islands (CI). S. pyogenes phage-like chromosomal islands (SpyCI) confer a complex mutator phenotype on their host. SpyCI integrate into the 5' end of DNA mismatch repair (MMR...
